I have been uploading files to my ICloud (which is taking forever) I have not synced them just dragging and dropping the relevant files I need and I have also turned off optimise mac storage. Once ICloud has uploaded them do they disappear from my Mac?

The design of iCloud Drive is to sync those files between your subscribed devices that are using the same Apple ID. It is not off device storage or a hard drive in the sky.

Partial answer is to go to System Preferences/iCloud (Apple ID for Catalina or later) and in the iCloud Drive line, click Optimize Mac Storage.
If you want the documents stored off the Mac, use another Cloud service like Dropbox.
