Is volume muted or set very low when headphones are plugged in? Speaker and headphone volume is handled independently. With headphones plugged in swipe straight down from the top Right corner of the screen and check the volume slider there in Control Center.

If volume is correctly set, try restarting the iPad: link-> Restart your iPad - Apple Support

If headphones still not working, check the jack for any debris that may be blocking the connection. Use a plastic or wooden toothpick and gently move around the jack. If nothing is in the jack, then it may have an issue and would need to be diangosed and repaired by Apple.
