You lock the screen by pressing and releasing the power button on the phone. The screen will go black. Do not press and hold, just press and release. However, the need to lock the screen is only important when doing the overnight backup. If you have opened the device and gone to iCloud backup and select Backup Now, the screen does not need to be locked during that time.
