Do I have to put the old SIM card from an iPhone 6s into the new iPhone SE in order for it to work??

Only if you want to make cellular voice calls with it. You will also need it to initially activate it. If you no longer have the SIM card, you can ask your cellular provider to give you another one.

Otherwise, if your cellular provider has already given you a new SIM card, just use it instead.
