Press and release the power button, or just let it auto-lock on its own. The need for the screen to be locked for the iCloud backup is only when you are wanting it to do the automatic backup. You can do a manual lockup with the screen unlocked while at the iCloud Backup setting. Momentarily pressing and releasing the power button will lock it.
