What if I set -up new phone with 'quick start' method and still need to transfer data from old phone from a back-up of old phone on my MacBook?

ONLY if there is something on the Mac backup that is not on the old phone.
If the Mac backup is very recent, then you are good to go as you are. Quick start will transfer everything.
You can't selectively restore parts of an old backup,
Your options are, provided you still have the old phone intact, Restore the new phone to factory, restore the backup from the Mac to the new phone, export the selective data in question to the Mac or a cloud drive, Restore the new phone to factory yet again then do the Quick start again.
It will need to be very important data to go through all that.
