I am missing a specific spreadsheet that used to be in a folder that no longer exists. How do I access a specific file (I know the file name) without being able to start Time Machine in the original folder?

You could enter Time Machine and go to the parent folder (or whatever folder is deepest in the hierarchy that still exists) of the deleted folder, and use spotlight (top right corner of the window) to type what you're looking for, and search back until you see it.
Assuming that you know the folder location, another method would be to open the most recent Time Machine snapshot that contains the file and manually traverse the file path using Finder.
