How do I lock my phone to back it up?

Press the button on the right side of the phone. That locks the phone until you unlock it with FaceID, TouchID or its passcode.

Also, unless you have Auto-lock set to Never it will lock on its own if you leave it idle for the time setting in Auto-lock.
