Could I disconnect an iMac backup disk?

Could I disconnect a backup disk? Will it be any difference when I reconnect it? Thank you.

Open Time Machine Preferences
Unlock the Padlock
Uncheck Backup Automatically
Lock the Padlock
Eject the drive
