I am not able set my favourite song as my ringtone after purchasing it from iTunes Store. I don’t see on my shoulder option

To set your ringtone as a song purchased from the iTunes Store, you'll want to follow the steps here: Use tones and ringtones with your iPhone, iPad, or iPod touch
The On My Shoulders option there in the screenshot is just a song that is selected. Are you trying to select the same song? Or a different song?
You will want to be sure you purchased a tone or ringtone and not just a song.
