is it possible to update OS on an iPhone SE 1st gen. if the iPhone has not been activated?

No, you have to activate the iPhone first.
After the iPhone is activated, you can update to iOS 15.x.x, see iPhone models compatible with iOS 15.2 – Apple Support (UK)
The iPhone will be update to iOS 15.3.1.
Go to: Settings -> General -> Software Update.
You need about 7 GB device storage.
If they are On, turn Off Hotspot & VPN.
