How do I make sure my screen is “locked” when backing up my storage? It’s taking much longer than it should.

If you are using the iCloud Backup Now option, you don't lock the screen.
If you want to automatically backup to iCloud while the device is charging, either lock the screen using the side button or do what I do and let it lock itself automatically based on the time chosen in Settings > Display & Brightness > Auto-Lock.
