Drive Letter? Windows this is not, and also Dropbox is not a drive or volume. Also, you can't use DropBox for a Time Machine back up.

How is it you want both of them to work?
I don't know if Time Machine will back up your Drop Box files, but I do know you cannot use your Drop Box (drive, folder, virtual storage location, snarfgargle, whatever you want to call it) as a backup destination in Time Machine.
It is suggested the user reads the Apple Support Documentation on using Time Machine Backup Utility which explains both the functionality of the Utility and what drives can be used
