Dropbox files are accessed in a folder on your Mac if you install Dropbox on the Mac, they can also be accessed through a web interface.

There is no way to assign Dropbox am drive letter on a Mac.

If you have the Dropbox files physically in that Dropbox-installed folder on your Mac, they are backed up by Time Machine. If the Dropbox files are stored only in the cloud (a Dropbox option), or if you only use the web based Dropbox interface, the file are not backed up by Time Machine.

Yep, and I never said anything WAS a drive nor drive letter.

Hard to argue that point:

How is it you want both of them to work?

I don't know if Time Machine will back up your Drop Box files, but I do know you cannot use your Drop Box (drive, folder, virtual storage location, snarfgargle, whatever you want to call it) as a backup destination in Time Machine.
