Not possible. Column labels are fixed as A, B, C... and we, as users, can not change the labels to numbers.
However, the COLUMN function in a Header Row will give the number of each column.
Perhaps that will help your aim.

If you just need the total number of columns and rows you could go to Format > Table and look at the numbers for Rows and Columns.
