You will need a lightning to usb adapter such as the Apple USB camera adapter and connect the card reader to it.

Depending on the card type, it may show up in Files app (memory cards with files), the Photos app (camera formatted memory cards), or in some app that can access other types of cards, like security cards, or credit cards for payment processing.
You cannot backup directly to. memory card, but you can copy items such as files, and photos to the memory card from the Photos app or the Files App.
