If I use transfer directly from previous iPhone will it also transfer everything I don’t have backed up on my iCloud from my old phone as well ?

Yes, the backup will transfer everything that is compatible with your new phone. In rare instances, some apps are no longer supported on newer models so those will not transfer. However, for the most part, everything should transfer. the only issues that I have run into in the past is that my mail accounts may need to be logged into again.

Although you will be transferring directly from the old phone to the new phone, it would be a good idea to keep backups up to date as well......just in case.
Everything will transfer over.
