Do you mean battery health (the overall ability of the battery to hold power) that you see in Settings>Battery>Battery Health? Or do you mean the current charge level?
Battery health starts to decline the moment the leaves the factory. That is an inexorable law of physics. Every time you drain and charge the battery, it's a little tiny bit closer to not being useful.
In order to put the phone into Standby Mode, it has to be plugged in. So, no, the current charge will not drop.
