Any edits I make to a screenshot upon taking it—that is, editing the screenshot as soon as it automatically appears on the screen immediately after taking it—are often discarded upon saving the 'edited' screenshot to to the Photos app. However this problem does not occur upon editing any photo—screenshot or otherwise—whilst working directly inside the Photos app.
iPad Air 3, iPadOS 16
There is bug in iPadOS15/16 that impacts markup/cropping of screenshots and Photos. This particular bug has been resolved in iPadOS 17.x.
For those users who have not yet updated from affected versions of iPadOS, there is a workaround...
First - save the cropped and/or marked-up image - at which point, when reviewed, the markup appears to have been lost. Saving another screenshot/image will make edits made to the previously saved image visible.
Interestingly, if an initial screenshot is triggered using the corner-swipe gesture with an Apple Pencil, the markup usually saves without needing to trigger a subsequent screenshot. If not already configured, you can configure the corner-swipe as follows:
Settings > General > Gestures >
Now that you have enabled the corner swipe, you can easily capture a screenshot with just your finger or Apple Pencil). Just swipe diagonally from the lower-left or lower-right corner of the screen towards the centre; this will immediately trigger the screenshot.
I'm very familiar with the corner-swipe function. Unless doing it specifically with the apple pencil makes a difference then this solution with regard corner-swipe is not universal, as I always use corner-swipe.
I wasn't aware of the first work-around you mentioned, but nonetheless it manifested as a further pain for me as it re-applied my initial edits to a screenshot onto a freshly edited screenshot, making a mess and necessitating starting over a second time.
Good that it's fixed in 17.
