Imac is Retina 4K 21.5-inch, 2019, it runs quite lot but I do have a lots memory in hard drive and in RAM. Wondering whether it's the problem of the slow spinning of my hard drive with 5400 rotation, in this case is it good to buy a SSD with 500GB ?

Potentially, yes ... but just replacing the Mac's system drive may not resolve the "slowness" issue. RAM will also be important. You would want your Mac to have, at least, 16 GB of RAM. If you are going through the trouble to replace the drive, I would also suggest that you consider getting a 1 TB SSD instead of 500 GB.
Yes you can startup and run you're 21.5" 2019 iMac from an external SSD.
