iTunes library sync with another Media folder?

I've done a LOT of work on my iTunes library and discovered the media folder it was linked to is the "wrong" media folder. If I change media folders in iTunes will the library "sync" with it? If not, can I make it do so? Much of the media folder(s) content is the same - only the one I want to use has much more media...

Can one of you iTunes heroes help me recover from my bone-headedness?
