Is it possible to upload photos directly from an external hard drive to iCloud? Mac storage isn't large enough so I can not transfer them from the external hard drive to my Mac then to iCloud.

Drag and drop. Don't use the Command key or they will disappear from the external. You could also use the import function in Photos but that is just adding them to your SSD on your Mac
