Is it possible to upload photos directly from an external hard drive to iCloud? Mac storage isn't large enough so I can not transfer them from the external hard drive to my Mac then to iCloud.

You can store your library on an external drive and sync to icloud. Move your Photos library to save space on your Mac - Apple Support Be sure to keep backups on a separate Time Machine disk
Drag and drop. Don't use the Command key or they will disappear from the external. You could also use the import function in Photos but that is just adding them to your SSD on your Mac
