I have messed up my password with a drugstore that I need to use often. How do I start over with them?

Nearly every major website has a link on their login screen that says "Forgot password". Visit your drugstore's website and look for that link near the username or password boxes. Use that link and follow the instructions to reset your password.
If you are referring to starting over with an entry in the Passwords app, simply find the entry in the password app and delete it. Then add the entry back with your new login information.
