I was copying some files to a USB Thumb Drive, I stopped and ejected the Thumb Drive. Now, when I insert the USB Thumb drive, it does not show up on the Desktop, or in "Locations" in the Finder sidebar? The file "Desktop" does not show in the Finder Sidebar either.

Launch Disk Utility and verify the physical drive appears on the left pane of Disk Utility. Within Disk Utility you may need to click "View" and select "Show All Devices" before the physical drives appear on the left pane of Disk Utility.
If the physical drive does not appear on the left pane of Disk Utility, then you have a hardware issue of some sort.
If the physical drive does appear on the left pane of Disk Utility, then run First Aid on the physical drive, then one the Container if it exists, otherwise run First Aid on the volume.

Within the Finder, click on Settings and select the "Sidebar" button and toggle the "Desktop" item until you get a check mark for the "Desktop" option.

Boot into Safe Mode according to How to use safe mode on your Mac and test to see if the problem persists. Reboot normally and text again.
Note: if you have a wireless keyboard with rechargeable batteries connect it with its charging cable before booting into Safe Mode. This makes it act as a wired keyboard as will assure a successful boot into Safe Mode.
1) Restart the iMac and re-Test.
2) Test the Thumb Drive in a different Port.
3) Test the Thumb Drive plugged into a different Mac or PC.
