I’m using MacOS 27.0.1 and mail on my Mac is not opening correctly. I’m getting a large rectangle box with a small blue square with a ? inside. My iPhone and iPad are operating fine. Is some setting messed up? I've read about going to Preferences (now Settings) but I'm not finding what they say should be indicated there.

That is an image that could not be downloaded in the message. There are several possible reasons, most commonly a server-side problem.
Perhaps a long shot:
Do you have a 3rd party VPN installed, even if you are not using it? If so, delete the VPN profile in Settings/General/VPN & Device Management/VPN, then restart your device. Don't just turn it off; delete the profile. Updates sometimes "break" VPN, and it needs to be removed. If that fixes it, you can try reinstalling VPN, but probably shouldn't.
