No, opening an app is meaningless. Go to System Preferences > Startup Disk > Does it show as an option to boot from?
If it does, choose it and restart.

Many Western Digital external drives won't boot a Mac with a PowerPC processor, but your MacBook is an Intel and you should be OK. See this WD support page for details:

As macjack said, the best test of whether a clone on an external drive is bootable is to try to boot from it!
