The backup for my iPhone 4 takes 15 seconds or so.
If the backup is taking a long time with iOS4, how many photos and/or videos are in your iPhone's Camera Roll?
If you only do the backup once a month and there is a problem that requires restoring your iPhone with iTunes, some of the data on your iPhone included with the backup will be lost. MacBook Pro 15" Intel Core 2 Duo/2.53GHz/4GB RAM/500GB HD, Mac OS X (10.6.4), iPod w/Video 30GB, iPhone 4 32GB

iTunes prefs, devices, and make sure "Prevent iPods, iPhones, from syncing automatically" is checked. I've been doing this for years, since the feature was added to iTunes a few years ago.17" MBP (C2D, 2.6Ghz), Mac OS X (10.6.4), MM (Intel, C2D, 1.6Ghz)

It still should not take very long - a minute or so at the most.
Our original iphone (8gb) backed up very quickly as does our 3gs ( 32 gb) and iphone 4 (32 gb).
