Is there a way to use British English, in terms of grammar and spelling with Mac OS X's spellcheck and autocorrect functions, but with a US Keyboard layout? Where # (Shift+3 on US layout) isn't changed into £ (Shift+3 on UK layout).

The keyboard layout is irrelevant to spellcheck. To set the spell check language you go to system prefs/language & text/text/spelling and set it to what you want there. Also you can go to system prefs/language & text/language and put British English at the top of the list (use the Edit button to add it if necessary). In any app you can also go to Edit > Spelling and Grammar > Show Spelling and Grammar and set the dictionary there (except Pages where you go to Inspector > Text > More > Language).

Oh. That did it. I never noticed the Spelling pull-down before.
Now I have my "colours" and "authorisations" working on my standard keyboard.
