iTunes running, your phone connected, right-click your phone in the left device pane, select: "Backup".
Your iPhone's backup is updated as the first step during the iTunes sync process, so sync your iPhone with iTunes immediately before doing so. Your iPhone's backup should also be updated as the first step of the firmware update process.

Contacts should be available on your computer. The iPhone is designed to sync contacts with a supported address book app on your computer so there should be no concern there.
Notes can be synced with a supported application on your computer as well.
Photos/videos in the Camera Roll can and should be imported by your computer as with any other digital camera. All photos/videos in your iPhone's Camera Roll should be available on your computer so no concern there either.
So does doing an update on the phone delete info on the phone? That was my main concern. Thanks for your help by the way.
Updating iPhone software does not touch any user data or settings, unless something goes wrong requiring a restore.
Thanks! It is still backing up, I guess this is normal since I have had the phone for over a year and never had done one.
