No, not if you use the same Mac App Store ID. Up to 5 Macs are free.
No! Go to the App store on your desktop, click purchased, then install Lion!

There is no limit of 5
It seems you are correct. From the Apple legal department:
ℹ to download, install, use and run for personal, non-commercial use, one (1) copy of the AppleSoftware directly on each Apple-branded computer running Mac OS X Snow Leopard or Mac OS XSnow Leopard Server (“Mac Computer”) that you own or control;
The glass ceiling of 5 installs probably comes from having the needed number of prerequisite Snow Leopard installs to upgrade from and Snow Leopard was sold as a single license and a Family Pack that could cover 5 machines. So, ... there's that. Maybe, the official Lion USB stick negates the SL requirement.
You can only authorize a maximum of 5 machines with your Apple ID. I believe authorization is required to use the App Store, which means (to me) an intended limit of 5 machines. Since Apple did not spell it out, and since there are ways to install that do not involve the App Store, this is likely to be a point of confusion for a while, but it certainly doesn't seem reasonable to me to install on more than 5 machines. Where else do you draw the line? 10? 15?
However, if more than 5 Macs were acquired by members of one family, with one person "controlling" those Macs, each with Snow Leopard originally installed, then one Lion acquisition covers them all.
So this may be the reason for the "limit of 5" rumor, but it is still unfounded.
Here is an excerpt from the Lion SLA; it is interesting because the license differs from the pre-installed OS vs. a purchase from the MAS; in any case, when purchased from the MAS, there is no limit - it very clearly states "on each Mac with SL you own or control". I believe the confusion about the maximum of 5 stems from the maximum of 5 computer allowed for iTunes songs.
SLA excerpt:
