When you press the sleep/wake button on the top of the iPhone, does the phone not react?
Also, have you checked the AutoLock setting to make sure it's not set to never go to sleep? You can check this setting by going to Settings>General>Auto-Lock.

Did you ever find a solution to this problem? I'm having it now, too. Just recently my phone stopped putting itself to sleep. It does go into sleep mode when I press the button on top of the phone, but it used to put itself to sleep after a couple of minutes without my having to do so. Now it does not. I have it set to go into "Auto-Lock" after three minutes.
I am also experiencing this problem. I have my phone set for 1 minute auto-lock and it has been working fine for last couple of years but not working since the iOS 6 upgrade.
Has anyone tried to restore the phone? or any other solution please?
Has anyone had a response to this? My iphone 4 has also stopped auto-sleeping, it has always been set to do this after 1 minute and has always worked perfectly but since ios6 it does not sleep when I am in an app ie mail, messages, phone etc. If I am on the home screen then it will sleep, and pressing the top button works.
I have tried to restore the phone to original settings (set up phone as a new), did try different settings in different modes with apps open, minimise or close and the 99 out of 100 times, the phone will stay awake which drains the battery a lot quicker.
Speaking of the battery, since the upgrade to iOS 6, its does not even last 12 hrs of a busy working day when I send/receive calls, check messages and emails (multiple accounts but with manual receive settings), browse the web on 3G and WiFi and use some apps. Have I gone too far in using the phone?
To all those having this Auto-Lock issue:
Are you using AssitiveTouch and iOS 6? If you are, there is a known nasty bug with the AssitiveTouch panel and the iPhone/iPad's ability to enter sleep mode (Auto-Lock).
The work-around is to NOT let the AssitiveTouch panel be the *last thing you press*, then wait for the AutoLock to kick in...it won't if AssitiveTouch is the last thing pressed.
Instead, simply touch the main screen somewhere or nudge it to the side a bit.

To get AutoLock to function normally, something other than the AssitiveTouch panel needs to be pressed. This can be a simple screen swipe.
Because of the additional motion (screen swipe or otherwise) required to get the screen to AutoLock correctly and not stay on indefinitely, I really doubt this is "by design". Seems like a nasty bug that will bite someone hard and kill their battery without them knowing it.
