I have downloaded io5 and the latest version of itunes. I have synced my ipad with the option checked for wifi sync. However, I cannot get the wifi sync button to ungrey on my ipad (or iphone for that matter) while connected to a pwer source. I can access the itunes library over home sharing. How do I make my computer available for sync?

iTunes is running on your computer correct.
If so, sync with the computer one more time. I think that's what I had to do. After the iOS update you have to sync with iTunes one last time - with your computer, then leave iTunes running to sync wirelessly.
Thanks...I have synced the dvice many times via the usb cable. Works perfectly. I have rebooted computers and devices. Itunes is open while trying to sync. I can home share so I know it is talking with itunes on the same netwrok. Yet when I initiate the sync from the device the option to 'sync now' is greyed and it says my specific computer name and that it is waiting for it to become available. I dont know what to do in itunes to make the computer ready to receive wifi syncs?
I have the exact same problem.
Not sure if it related but iTunes keeps opening after it has been closed, both of these problems are happening to me and a co-worker.
One thing that maybe you and JasonVRN have not done yet is to select Sync with this iPad over Wifi in the iTunes summary tab. You have to do this while the iPad is connected to the computer and while iTunes is running. Then you should be good to go.
@JasonVRN - do you have iTunes selected to open when the iPad is connected? That option is right above the WiFi sync option in the photo below. I do not use that option, but that may be the reason it keeps opening. I really don't know how that option works now with WiFi sync. I never used it to begin with - ever. I always open iTunes on my own and I never did auto sync either. I always initiate the sync myself.

Go to System Preferences> Energy Saver and you will see a checkbox marked ''Wake for Network Access''. If unchecked, check this box and exit (this is for when connected to mains, not battery)
This allows the system to enable WiFi sync.

Uk-max i can't find this option n my iPad or iTunes.
It must be a problem with iTunes as I cant sync my iPhone wirelessly either.
JasonVRN - I had the same issue with trying to sync my iPhone via wifi and found out is that one needs to have the minimum OS installed on their computer in order to section that Demo showed in his response. If you don't have the minimum required OS, then it will not show.
I have a Powerbook G4, but can only go as high as Leopard as an OS on my system. I think you need Snow Leopard and beyond to be able to sync over wifi.
The systems preferences setting is on the mac, not the pad? Aha. I never even knew that existed. Mine would not sync if the computer was 'asleep'. Now i know why. So turn wifi sync on both on the pad and the computer, leave i tunes active on the mac, and set system preference to wake the mac up. Got it. I think.

I have ticked the sync this iPad / iPhone over wifi option in iTunes on both devices
Neither the iPhone or the iPad 'Sync Now' option is available when only connected via wifi.
I understood you but I don't know what else to tell you to try.
Sync one more time with iTunes - non WiFi
Turn on WiFi sync in iTunes while the iPad is still connected to your PC.
Click on Apply in the lower right corner.
Eject the iPad.
Go to Settings>General>iTunes W-Fi Sync>Sync Now
That option I described is on your Mac, which I now see is actually your PC.
Sorry, not a Windows man - know nothing about them. I would look at that end though, rather than the ios5 device. There will most likely be something similar in Windows that is preventing connection.
Thanks all for your thoughts on the orginal post of mine. I truly believe it is a glitch somehow in the new itunes or io5 as others are having the same difficulty. I read in other posts that the device just 'appears' in itunes after you have gone through the whole process of checking the box and updating. Mine simply never appears. I have given up for the moment as I can easily still sync with the cable and wait for an update from apple to see if that helps. I am just totally puzzled since I can easily home share with my itunes and iphone and ipad 2. It would seem to me those have to be on the same wifi network, etc and I have no trouble with getting them to talk to one another. I have no idea why my wifi sync is simply not working. Thanks...
I have had the same problem with my ipad and iphone reporting that my computer running itunes was unvailable. I resolved it by changing my wifi router security encryption from WPA-PSK - WPA2-PSK TKIP - AES to WPA2-PSK AES only. The mixed mode seemed to be the problem, and I was also able to synchronise with WPA-PSK TKIP. This allowed me to sync over wifi successfully and I hope it assists in your problem.

Always looking for new ideas...switched security and rebooted everything and unfortunately it sill didn't work. Just don't know why I can easily home share but not wifi sync. It is just so puzzling to me...

Use Demo's screenshot above as a reference - you need to first enable Wi-Fi sync from iTunes. (You haven't indicated that this had been done.) Plug your iPad in with the cable - then in your device's menu, you'll see "Sync with this iPad over Wi-Fi" as an option in Summary page under Options. Only after that, you'll see sync over Wi-Fi on iPad. (Home Sharing and Wi-Fi Sync are separate options, the former is enabled from the device, the latter needs to be enabled from iTunes.)
