Is there a chance Apple is giving this back? This was something I was able to do before upgrading to iOS5. It was actually the main reason I bought an iPad2. After the upgrade what happens was when I click to open a video file, it opens and closes back. It was like those dissolve effect used in photos.

You can still view videos on the iPad in exactly the same way as before. Apple hasn't taken anything away.
Sounds like something is up with yours. Have you tried a reset?
My question to you is - Were you using Home Sharing on the iMac and watching the videos that way? If the video is downloaded or synced to the iPad, what does having the computer running have to do with this issue at all unless you are talking about home sharing?

Actually, if I remember right, after upgrading to iOS5 home sharing popped out with the upgrade so I had to use it. It's either that or go to iTunes store. Videos were synced to the iPad from iTunes which was done before the upgrade and I had no problem watching my videos. Then came iOS5 when I couldn't watch anymore with the computer off.

Try this reset ....
Reset the iPad by holding down on the sleep and home buttons at the same time for about 10-15 seconds until the Apple Logo appears - ignore the red slider - let go of the buttons.
If that doesn't help - try quitting the videos app and restart the iPad.
Go to the home screen first by tapping the home button. Quit/close open apps by double tapping the home button and the task bar will appear with all of you recent/open apps displayed at the bottom. Tap and hold down on any app icon until it begins to wiggle. Tap the minus (-) sign in the upper left corner to close the apps. Restart the iPad. Restart the iPad by holding down on the sleep button until the red slider appears and then slide to shut off. To power up hold the sleep button until the Apple logo appears and let go of the button.
No need to completely reset content or restore the device yet.
I just tried this and nothing happens. My videos only shows up when the computer's on.

That would indicate that you are streaming the content from your computer to your iPad. If you wish to watch content when not on your computer's local network (like on a trip) you need to put it onto your iPad.
To add content (music, videos etc) to your iPad: Connect your iPad to your managing computer and use iTunes to select what you want to transfer it. Details are in the user's manual.
Note: the users manual can be found on the device (bottom of Safai bookmarks), viewed online from the Apple support section.
I did mention that's what I did in one of my replies here to this discussion. And I did this before the upgrade to iOS5 and I was able to watch videos with no problems. After the upgrade, you're only given two choices, Store (iTunes) or your library (this disappears when you log off your computer). You're not given a choice to select what you have in your iPad. It's clear to me that this was written off by the upgrade.

Yes. This was were I used to be able to access the videos in my iPad. Not anymore after the upgrade. I actually tried republishing the videos in iTunes. Computer still needs to be on.
You state "republishing"... are the videos you are trying to watch ones that:
a) you purchased from the iTunes Store?
b) you have told iTunes to sync to your iPad (using setting in iTunes then clicking the Sync/Apply button) and transferred?
c) items that you have previously published to MobileMe?
If the answer is C then I'm guessing you upgraded to iCloud and that is the reason. Moving to iCloud kills: iWeb, iDisk, photo and video storage and hosting that you used to have with MobileMe. Ars technica article on the switch pros and cons.
If you did not buy an item from the iTunes Store then it must be manually configured (or all videos) in iTunes for a sync with iTunes on your computer.
I didn't buy any of my videos from iTunes. These are music videos I did myself. The term republishing is one used in iMovies when you want to share your iMovie projects. This is what I did and did these before iOS5 and it worked. After the iOS5 upgrade, when I select a video to open/play, it will close back. Similar to a dissolve effect used in photos.
I just want to let you guys know that I decided to call Apple support. The lady who answered my call sounded pretty confident that their tech support team can downgrade my iPad to iOS4 so she transferred me over to them. The guy at tech support told me they don't do that. And after I've told him my problem, he was really willing to help me but he had to check with his supervisor and you know what happens after that was he told me I have to sign-up with Apple Care for $29 for one phone tech support or $79 for 2 yrs. cause my warranty already expired. But I said it was iOS5 that created the problem for my iPad. I was really happy with it before that upgrade. Is this something that Apple does that we have to live with? They create the problem and we have to pay them to fix it? This is not fair!

I still don't understand exactly what the problem you are having is.
Apple have not removed the ability to view videos on the iPad, synced from iTunes. Nothing has changed in that respect. Your Mac does not have to be on to view videos on the iPad. I take my iPad on trips with me, away from my Mac and videos I have put on it still play.
I feel you have missed out some vital information about exactly what you are doing, as importing videos in iTunes, syncing them to the iPad, and then watching them in the Video app has not changed in iOS5.
There has to be something that is getting "lost in the translation" here from you. Like Julian, I can watch any video that I have synced to my iPad whether my Mac is running or not - even if I am 100 miles away from the Mac. The Mac being turned on has absolutely no bearing on the ability to watch synced videos on the iPad. Either we are misunderstanding your situation, or you are not explaining it properly.
My suggestion to you is - make an appointment with the Genius Bar at an Apple Store, and take your iPad to them and then let them take you through the process of how to watch a video on the iPad. Explaining to them in person - not over the phone.
