There are many ways to do it. If you ever forget the keyboard shortcuts and are in a hurry, then you simply can activate Preview and from the "File"-menu select

File -> Take Screenshot -> From selection / Window /Entire Screen

Your Screenshot will open in Preview as a new image that you can save or modify.

If you need to take a screenshot of windows or panels that will disappear if you switch to the Finder or Preview to take the screen shot, you can use the "Grab" utility, from "Applications"->"Utilities"->"Grab"

Grab can take timed screen shots: From "Grab" start a timer, that will give you ten seconds to arrange your Desktop with all panels, drop down menus, or pop up menus from within the Application you are recording. That should even work with windows on the Parallels Desktop.

I am not sure however if "Grab" came with Lion or is still on my System as a left over from previous Systems.

These features are also listed under "Services" in the system pulldown menu.

So if I'm on Safari, I select Safari from the menu bar and then Services.

My favorite one, which wasn't given here yet, is window capture.

To just take a picture of the window you are on, hit shft+cmd+4 and then spacebar. This will make your cursor a camera and will highlight the window it is over. when you click the mouse, it'll take a picture of the window it is highlighting.

Ok, Maybe I'm stupid, but when i use cmd shift 4 and draw the window I hear the picture sound but can't find the picture . . . pasting does not work (the previous item copied gets pasted). Where is the image?

If the screenshot is not placed on your desktop, then perhaps you are accidentally pressing Command-Control-Shift-4

Adding Control to the key combo will place the resulting screenshot on the Clipboard rather than create an image file and place it on the desktop.
