Drive letters aren't used on a Mac. If you're interacting with it from a normal application, it's named Macintosh HD by default. If you're using the Terminal or other command line tools, it's /.

No. It's Macintosh HD; if it doesn't appear, change the settings in the Sidebar tab of the Finder's preferences.
