Is it possible to auto refresh "Find My iPhone". This would allow me to use it as a tracking application and help my wife NOT get lost again!

Find my Friends would be a better solution.
You might want to take a look at the new Find Friends app that launcehd with iOS 5.

Yes. With the location dispalyed on the map, click the Refresh arrow/circle in the upper left box to the right of My Devices.
And as KT said, Find my Friends might work better for you.
Or maybe a Garmin Nuvi for your wife? 😉
