remove item from startup disk os 10.6.8

My HD is 498 gb of capacity? Startup disk is full, delete items. I emptied trash. How do I remove items from startup disk. I want to upgrade to lion.

MacBook Pro, Mac OS X (10.6.8), Just convert from PC
