Grey screen annoying me, any help please?

Ok so i have a 20" Intel Imac from 2007. It has been working perfectly and without flaws for all these years. But since like a month ago it has been booting to a grey screen and thats it, it just sits there for like 45 mins and then shows a folder with a question mark in the middle. I have tried almost every "booting trick" i have read about, except for the disc booting because i don't know how it will affect all my saved files i need for work. PLEASE HELP!! I NEED MY PATIENTS HISTORY FILES,,,,,,,,

iMac, iOS 5.0.1

Safe booting will not affect your files.

No it won't. Have you moved any part of the system, or any applications, to a different location where the start-up sequence can't find them?

Two other possible solutions:

Run the latest 10.6.8 combo updater again.

Or perform an archive and install:
