Can I use Apple TV with my MacBookPro to play a DVD on it and display it on a large screen?

No, you cannot do that.
Why can't you stream from the computer to the Apple TV?
Hi Steve, you can stream from the computer to the Apple TV, however you can only stream content which resides in iTunes, not from the DVD player.
I understand that this might be possible with mountain lion , whilst additional airplay features have been announced there has been no mention whether there will be any hardware restrictions that apply.
Gotcha. So the OP just needs to put the DVD content into iTunes. I've streamed directly from my computer and from an iPad using direct connections to the TV, but not using Apple TV, hence my question. I thought you should be able to stream anything played on the computer to the ATV via Home Sharing.
Assuming it's a home-made or nonprotected DVD then yes, it needs to be converted and the resulting file placed in iTunes.

So effectively, there's no other way to play a DVD movie using, say, VLC, and stream it to the TV other than a direct connection to the TV via HDMI cable/adaptor or the Apple VGA Connection Kit?

Based on the information, I have to upload my DVD (home made) to iTune, then connect my MacBookPro and Apple TV via HDMI connector to a monitor for iTune to play it.If this is correct, can I have few monitors connected at the same time to play it. I would like to show my DVD to an audience as a presentation. There would be more than 100 people present. Or do you know any other approach to solve this issue for me?

There is, but the video/audio sync seems to lag at times. AirParrot allows you to AirPlay your Mac to the Apple TV. You can extend the desktop, or mirror.
Thank you. So, there's a possibility except for a video/audio sync lag. Too bad. Then how did Apple (Steve) etc use their equipment when they gave a conference on new products and showed it to a large audience?
Mountain Lion is said to bring native Mac miroring, will have to wait and see.
