I never have but you should always back up first.
You shouldn't, but possible.
Photos on your iPhone that were transferred from your computer should remain on your computer.
Photos in the Camera Roll can and should be imported by your computer as with any other digital camera. Not a good idea to depend on your iPhone alone for these photos which can be lost or stolen or damaged beyond repair accidentally.

Included with your iPhone's backup but so is a lot of other data.
There is no guarantee your iPhone's backup won't have a problem in regards to this data.
Do you have to manually save photos and video from iphone somewhere? Or does it back up automatically when you plug it into your computer?
It doesn't back up automatically but when it does backup, photos and videos are backed up. That being said, you should regularly copy or move photos and videos taken by your iPhone to your computer in case your iPhone crashes.

Photos/videos in the Camera Roll are included with the iPhone's backup. If you choose to have iTunes handle the backup, the backup is updated as the first step during the iTunes sync process. The other option is having the backup done wirelessly with an iCloud account.
Thank you for quick responses... When I plug my phone in it always asks me to import photos to iphoto but I reject it because I ran out of space in iphoto previously. But are you saying that through itunes, it has saved everything somewhere?
I don't know if I have icloud- I can't remember.
My phone is frozen with the itunes icon and a usb plugin pointing to the itunes icon...
What do you mean you ran out of space in iPhoto? iPhoto is not a place. Do you mean your hard drive is full? You need to address that before you do anything else. If your drive does not have enough space to extract the photos to iPhoto, then it does not have enough space to successfully back up your phone.
If you have the iTunes logo/USB plug on your screen, the phone is in recovery mode. Anything that was on it is gone and it will need to be restored.
Yes, I guess it was my hard drive that ran out of space because I kept so many videos and photos. The apple guy at the store said I need to save them on an external device rather than put them in iphoto. Now I'm not sure if that is correct or not but I've been afraid to keep them there.
