can I use malware update if I'm not running OSX lion?
can the malware app be used if OSX 10.5.8 is on my computer?

Only 10.6 and up are covered by the updates. There is no remover app, the update itself does the removing. You'll need to use other tools to detect and remove an infection, and will have to disable Java.
