Connect the iPhone to a computer and launch iTunes.
Select the iPhone under Devices left side of the iTunes store.
From the Summary tab in the menu click Check for Update

If you have a 3G instead, and I only say this because most folks are clueless what model they have, 4.2.1 is the latest version available.
