Connect it to iTunes, click on the name of your iPhone in the left frame under DEVICES, move to the right to the Summary tab, click on the Update button.

Are you sure you have a 3Gs and not a 3G? The 3G cannot update past 4.2.1.

If iTunes doesn't automatically prompt you to update, do the following:
Locate and click on the name of your iPhone under the "DEVICES" header on the left in iTunes. Click on Check for Updates. Afterwards, an update button should appear.

Another possibility is that it says iOS 4.2.1 is the current version. If it says that, then you have an iPhone 3G and not an iPhone 3GS, and an iPhone 3G cannot be updated to iOS 5.
