As it may take 22 hours, will the back up continue when the display goes to sleep?

Display should be fine.. but you haven't said what kind of Mac your talking about.

Must be a huge backup. Using an Ethernet connection between your Mac and the Time Capsule, you should expect a copy speed of about 25-30 GB per hour.
Bets are off if you are using wireless.....it will probably be at least 4-5 times longer, maybe more....along with a significantly higher chance of something going wrong.
is it ok to let the display go to sleep during the first back up with time capsule?
