Is it possible to have a 20-sec video clip intro for a subtitle? The subtitle consists of a jpeg picture with audio background
I have been unable to add anything - video or audio clip - to the designated area when constructing the subtitle contents

I could be wrong, but I don't believe iDVD has that capability; the subtitles are for text, not a movie-in-movie (that's about what you want to do if I understood correctly). Depending on which version of iMovie you're using, you may be able to construct it there (the entire thing, import the background and then use some third party plug in to allow to add a movie in a corner), export it, and then drop it into the background area of the iDVD menu/theme.
Do you want this 20 sec. video clip to play before the ???? What do you have in the iDVD project? A slideshow of stills created in iDVD or a movie/video file?
If it's the slideshow of stills you can add a video clip at the beginning of the slideshow. iDVD 7 not allows video clips to be added to slideshows:
If you want the video clip to play immediately when you put the disk in a DVD player and then fall out to your menu just add it as movie to the menu and then, in the Map mode, drag that clip into the Auto Play bin.
The clip will play and then fade into the main menu.
If those are not what you're trying to do describe in detail exactly what it is.
OT
The clip is not for when first putting the CD in - that's already set up - but I'd like it to play similarly when a separate SubTitle is selected on the DVD.
Ideal scenario is that the SubTitle is selected, the clip starts and then rolls into a, not a slideshow, but single PDF with music background. Ive been unable to place anything in the INTRO box; everything else - the PDF still and music background - is set-up, but I can't drag the video in the intro box, even when the intro is checked (the video is already in itunes).

That sounds like something for iMovie or FCE.
