I've just downloaded iTunes 10 and now I've lost all sound. Anyone know what I did wrong?

you may have inadvertently connected to airplay speakers in iTunes? This will make it seem like everything is playing fine, but you don't have sound. check in the lower right corner of iTunes to make sure it says "computer" not whatever your (presumed) airplay device might be.
If that is not the case:
You may want to check out Audio Midi setup in Applications/Utilities to see what you have your sound output to as well.

Thanks very much for you response. Ultimately I went to System Preferences - Sound and hilighted internal speakers. I think sound was going through iParrot (for wireless TV connection). Thanks.
I've had some other issues with airparrot and lion. ultimately a drag and drop issue. if you find that your items are sticking to your cursor when you are dragging and dropping, hold esc key and click, it should snap back to where you were dragging. Then go to the apple and put your computer to sleep and then wake it back up.
I don't use airparrot because of this bug with Lion. Maybe they fixed it?
I've had that issue of sticking files. It has only happened intermittently and the logitech mouse I'm using seems unstick it after some manic clicking. I thought the issue predated Airparrot, but I could be wrong.
