I have couple of .mov format files provided by my client. I am not able to play the file in QuickTime player 10.1 in Max OS X v10.7 Lion. Please give me a good suggestion/solution to solve this issue. I expecting your valuable support/help.

Try to install another video player. VLC its free and plays everything: Files, Discs, Webcams, Devices and Streams.
Plays most codecs with no codec packs needed:
MPEG-2, DivX, H.264, MKV, WebM, WMV, MP3, MOV, and much more...

Though I installed perian in mac OS X v10.7.1 Lion still the QuickTime 10.1 player does not play .mov file. Do I need to do anything additional?
Please let me know any other options to try again.

There may be something wrong with his files. Do you have any other .mov and do they open and run correctly?
All of my .mov functions correctly. If your own files work, I'd think that something got corrupted in the transfer.
Do you get any specific error messages (quote them exactly) that might help us figure out why thy are not playing?
I am having the exact same issue.
I have tried mov files from mutiple sources such as my iphone, itunes and videos converted from my HD Camcorder and none of them will play in quicktime x, i can force them to play in quicktime 7, but for some reason not in x.
I am also unable tp play any of the videos on the apple homepage, or for that matter play or download movie trailer from the apple trailers page.
If I use Firefox, i can play the embedded videos from apple (both homepage and trailers) however i cannot choose the download option for the movie trailers.
I also cannot ge the mov files to play in itunes either and this includes videos download from itunes on windows 7 pc rather than mac.

Soupladel - It is unlikely this is the exact same issue. I suggest you post in a new topic so we don't muddle helping you with the help we are providing the originator of this topic.
Try playing it with VLC. I know this isn't what you want to do but we're trying to figure out if anything will play these. They could be corrupt or improperly made. What system is your client running?
Has anyone figured out a solution for this yet? I have seen countless forums now regarding the same issue with QuickTime Player 10.1, so there is clearly a problem with this version of QuickTime. It is just frustrating that I have this brand new up-to-date computer and am having issues 😟.
I am in agreement with MelanieChantel -- any progress on this question? I just updated my systems to 10.6 (yes, I'm a laggard), and some .mov files I can open with Quicktime 10.0, some I cannot...but ALL can be opened with Quicktime 7 (and this is Quicktime Pro I have on my MacbookPro 2008). Just bizarre -- at least I thought to check with my QT 7.
I recommend that you download and install the following Free player, which will solve the problem:
