Can I combine factory installed 2 GB in two slots with 8GB RAM in the now two empty slots? That would be 2+2+8+8 =20 GB RAM! Would that work?
My iMac 27" is a Model ID iMac11,3 IntelCore i5 2.8 Ghz I purchased it in September 2010.

Here is a link to the specs of your Mac. Apple officially supports up to 4 GB in each slot - OWC has tested it with 8 GB, so it'll be your decision.
It's really the question about mixing the sizes in different slots. When I get finished, I'll have this configuration:
Slot 1: 2 GB
Slot 2: 2 GB
Slot 3: 8 GB
Slot 4: 8 GB
Will that work, assuming they are all Mac compatible RAM sticks?

They need to match horizontally: 2 2GB next to one another and 2 x 8 gb next to one another.

You're welcome and don't worry about it - glad I was able to help. Remember that OWC has tested it with 8 GB modules, but officially only 4 GB modules are supported in each slot, so...... just in case you need to take yours in for any kind of service/repair, I'd suggest removing your 8 GB modules so whatever problem cannot be blamed on the "unsupported" amount of RAM which may make warranty coverage questionable.
That's good advice on removing it when getting service. I did just this setup - adding 2 fours from OWC and it's great to have this much memory - it's also on an i5 iMac.
