No. The update will delete it and you won't have to do it manually
Still cannot connect to the Internet. I'm communicating to you via a PC. But I am running MacScan in the background. Is that a reason why I still cannot connect?

MacScan won't prevent you from connecting to the internet. Neither does FlashBack. As long as you have the latest Spyware Definitions dated July 6, MacScan should tell you if you contracted the DNSChanger Trojan which will now prevent you from accessing the internet.
