Is there a big difference between HD and SD films purchased from the iTunes store on the latest iPod touch?

Yes. About the same difference as SD and HD on a TV.

One of the only major differences is the resolution the video is formatted with. SD versions are formatted at 480p, while SD versions are either set at 720p or 1080p.
