Is this a 1st or 2nd gen shuffle or a 3rd or 4th gen shuffle? 4th gen is the current shuffle. This article about syncing the shuffle shows the four types.

In either case, you can do a Restore on the shuffle in iTunes, which will erase the shuffle and give you a "start over," This is done by select the shuffle in the iTunes sidebar, under devices, and going to the Settings or Summary tab (depends on shuffle model) to the right. The Restore button is there.

However, you may be asking how to delete songs from a shuffle. That depends on which shuffle model you have.

1st or 2nd gen - Select the shuffle in the iTunes sidebar, under DEVICES. Over to the right, go to the Contents tab. You should see a list of songs on the shuffle. Select the song you want to delete and hit the Delete key. You can select multiple songs at the same time. If you want to delete ALL songs, click on any song and select Edit -> Select All from the menu bar, then hit the Delete key.

3rd or 4th gen - If you added songs to the shuffle manually (by dragging songs to the shuffle in iTunes), select the shuffle in the iTunes sidebar, under DEVICES. Click on the little triangle next to the shuffle to "drop down" the content categories, indented under the shuffle. Click on Music there (under the shuffle). To the right, you should see a list of songs on the shuffle. You can delete songs from this list (as described above for 1st or 2nd gen shuffle).

If the shuffle (3rd or 4th gen) is set to sync automatically, or you want to set it up to sync automatically (which is more convenient in my opinion), you need to set up the iPod's Music tab in iTunes. This is done by selecting the shuffle in the iTunes sidebar, under DEVICES. Over to the right, go the Music tab. On this screen, you define how you want iTunes the sync the shuffle automatically, every time you connect it. You can post back if you have more questions about doing it this way. That article linked above also provides more details.
